Overview
Eloralintide (LY3841136) is an investigational amylin analogue. Laboratory assays found preferential activation of human AMY1 receptors over the tested calcitonin and AMY3 receptors. Published randomized human studies evaluated subcutaneous administration for weight-management research. Standalone eloralintide results must not be replaced by combination-treatment outcomes.
Research Summary
The standalone Phase 2 trial randomized 263 participants at 46 U.S. centres for 48 weeks, excluding type 2 diabetes. Under the efficacy estimand, mean body-weight changes were −9%, −12%, −18%, −20%, −20% and −16% for the 1 mg, 3 mg, 6 mg, 9 mg, 6-to-9 mg and 3-to-9 mg regimens, respectively, versus −0.4% with placebo. Nausea rates in that same regimen order were 11%, 13%, 64%, 33%, 54% and 25%, versus 14% with placebo; fatigue rates were 0%, 13%, 29%, 43%, 46% and 21%, versus 12%. These are regimen-specific trial observations, not dosing instructions or long-term safety estimates. An earlier publication reports in-vitro receptor studies, animal research and a randomized single-dose study in 48 healthy participants; these evidence layers remain separate. Eloralintide plus tirzepatide outcomes are not used as monotherapy evidence.
