At a Glance

An NAD+ precursor with human biomarker and tolerability research; clinical benefits for aging or disease are unestablished.

Plain English

NR studies can show how the compound is absorbed and affects NAD-related measurements. Those results do not yet show that it prevents disease, improves cognition, or extends life.

Overview

Nicotinamide riboside (NR) is a vitamin B3-related NAD+ precursor. Selected human studies report tolerability and NAD-related biomarker findings in their studied participants; they do not establish benefit for longevity, cognition, cardiovascular disease, or metabolic disease. NCT02950441 is a specific registry record with unknown status and no posted results, so it is not efficacy evidence.

Research Summary

Trammell et al. reported oral NR bioavailability in a human study, and Conze et al. reported tolerability and NAD-related biomarker findings in healthy middle-aged and older adults. Those findings do not prove prevention or treatment of disease, cognitive benefit, or longevity benefit. NCT02950441 has no posted results and is not used as standalone efficacy evidence.

Safety & Risks

Serious Risks

  • Long-term clinical safety and disease-outcome benefit remain unestablished.
  • NAD-related biomarker changes do not by themselves demonstrate a health or longevity benefit.
